The Almendares River watershed covers a large portion of Havana, Cuba and is centrally important to both recreational and other activities in the region. In order to assess current water quality conditions prior to planned remediation efforts, the spatial distribution of six heavy metals and other compounds were determined in river sediments at fifteen sampling stations in the watershed. Metal concentrations in sediments ranged from 86.1 to 708.8 for Zn, 39.3 to 189.0 for Pb, 71.6 to 420.8 for Cu, 84.4 to 209.7 Cr, 1.5 to 23.4 for Co, and 1.0 to 4.3 for Cd microg/g dry weight sediment. Calculated enrichment factors (EF; measured metal versus background mineral conditions) were almost always greater than 1.0, suggesting significant anthropogenic impact on metal levels in the river. The highest EF values were seen immediately below Cotorro (EF>10 for Pb, Cu, and Cd), a suburban town that has an active secondary smelter, and below the largest municipal landfill in Havana (EF>10 for Pb, Cu, Cd, and Zn). Further, three sampling stations had multiple metals at concentrations higher than probable effects concentrations (PEC), implying possible local ecotoxicological impacts. Finally, sequential extractions of the sediments indicated that heavy metals were largely associated with the organic fraction, and it was estimated that up to 62% of metals in the sediments would be susceptible to release back into the water column if hydraulic or other changes occurred in the river. These data are being used to prioritize decisions related to the remediation of the river system.  相似文献

Advances in microbial source tracking (MST) have largely been driven by the need to comply with water quality standards based on traditional indicator bacteria. Recently, a number of culture-independent, and library-independent methods based on polymerase chain reaction (PCR) have been gaining popularity among source trackers. However, only a limited number of these methods have been successfully used in field applications, primarily due to the fact that many of them are still being developed. In this critical outlook, we examine different viewpoints associated with the practical use of MST to identify critical research gaps, propose a priority-based timeline to address them, and outline emerging technologies that will likely impact the future of source tracking. We propose that it is necessary to consider each of these aspects in order to advance towards a unifying framework in source identification, so that fecal pollution monitoring can be reliably used for comprehensive environmental microbial monitoring, to develop risk assessment models, and to implement and validate adequate management practices.  相似文献

An experimental study was performed to evaluate the bond strength between two concrete layers of different ages, considering different mixtures of added concrete, with different strengths. The specimens first had the roughness of the substrate surface increased by sand blasting. Later, the new concrete was added. Afterwards, slant shear tests were performed to quantify the bond strength in shear. These tests indicated that increasing the compressive strength of the added concrete relative to the compressive strength of the substrate concrete improves the bond strength and changes the rupture mode from adhesive to monolithic. A finite element analysis showed that, increasing the difference between the compressive strengths of the added concrete and the substrate concrete, higher values of normal stress are present in the interface, for the same level of shear stress. The study presented in this paper reveals that the added concrete, normally with higher compressive strength than the substrate concrete, may possibly have an influence on concrete-to-concrete bond strength. Therefore, design of shear at the interface between concrete cast at different ages, according to Eurocode 2, could be improved, since here only the lowest strength concrete is considered.  相似文献

In the first part of this paper, an analysis of the data collected during the American Association of State Highway Officials (AASHO) road test, based on probabilistic duration modeling techniques, is presented. Duration techniques enable the stochastic nature of pavement failure time to be evaluated as well as censored data to be incorporated in the statistical estimation of the model parameters. The second part of this paper presents the use of economic optimization principles for determining the optimal design of flexible pavements. We study the effect of deterioration model accuracy on optimal design and lifecycle costs by comparing three models. The first is a simple regression model developed by the AASHO, which forms the basis of design standards in use today. The second is a regression model that was developed with the same AASHO data set, but that includes a correction for data censoring. The third model is the probabilistic model developed in the first part of this paper. The results show that the AASHO model, when used as an input to lifecycle cost minimization, produces a pavement structural number that is lower than that produced by using the other two deterioration models. This results in shorter pavement lives and higher costs due to more frequent resurfacing. The savings in lifecycle cost accrued by using optimal structural number are shown to be quite significant, offering a sound basis for revising current design practices.  相似文献

Dearomatization levels during electrochemical oxidation of p-methoxyphenol (PMP) and p-nitrophenol (PNP) have been determined through UV-Vis spectroscopy using solid phase extraction (UV-Vis/SPE). The results show that the method is satisfactory to determine the ratio between aromatic compounds and aliphatic acids and reaction kinetics parameters during treatment of wastewater, in agreement with results obtained from numerical deconvolution of UV-Vis spectra. Analysis of solutions obtained from electrolysis of substituted phenols on antimony-doped tin oxide (SnO2Sb) showed that an electron acceptor substituting group favored the aromatic ring opening reaction, preventing formation of intermediate quinone during oxidation.  相似文献

The objective of this study was the quality loss inhibition of hake (Merluccius merluccius) during the frozen storage. For it, the effect of a previous high-pressure (HP) treatment (150–450 MPa for 2 min) was analysed throughout a 5-month storage at ? 10 °C. Quality changes were monitored by complementary chemical analyses. Inhibition (p < 0.05) of dimethylamine (DMA), free fatty acid (FFA), formaldehyde (FA), trimethylamine, total volatile amine and fluorescent compound (tertiary lipid oxidation compound) formation was concluded by previous pressure treatment according to the one-way ANOVA analysis. On the contrary, no effect (p > 0.05) on the K value, polyene index and formation of peroxides and thiobarbituric acid reactive substances was achieved. Additionally, a multifactor ANOVA test (pressure and frozen storage time effects; i.e. comparison among HP treatments) showed an inhibitory effect (p < 0.015) on DMA and FFA formation, this effect increasing with pressure level applied. This inhibitory effect on the formation of such molecules related to quality loss can be explained on the basis of the damage caused to different kinds of enzymes such as trimethylamine oxide demethylase, lipases and phospholipases, so that their activity during the subsequent frozen storage would decrease. The work here presented provides for the first time information concerning the employment of HP technology to inhibit the DMA, FA and FFA formation during the frozen storage of hake. Further research focussed on commercial frozen conditions (? 18 °C) and including sensory and nutritional aspects is foreseen.  相似文献

Previous earthquakes, such as the 2010 Maule earthquake in Chile, have demonstrated the need to establish suitable predictors of compressive or tensile strains in concrete or steel in reinforced concrete shear walls, which can provide limit states or confinement requirements. Slender walls are commonly controlled by flexural deformations that can be divided into elastic and inelastic components. This study provides calibrated expressions for the elastic and inelastic components of flexural deformations using a fiber model for slender walls. These expressions are obtained for rectangular and T‐shaped walls. The elastic component is dependent on the axial load and the boundary steel reinforcement ratio. The impact of wall coupling is investigated, which requires a correction for the elastic component. The investigation of the inelastic component is based on a plastic hinge model, in which the length of the plastic hinge is a function of the lateral inelastic drift of the wall among other parameters. The traditional linear inelastic curvature distribution over the wall height is also modified for cases with steel reinforcement with a long yield plateau or low strain hardening, which results in a larger curvature at the wall base. This article analyzes an important dimension in which the organization of the projects performed by the same firm can differ: the insourcing or outsourcing of an activity that needs to be undertaken in each of the different projects. Analyzing the variability of a firm's insourcing or outsourcing decision across its projects gives us a better understanding of the firm's decision-making process in terms of the stability of its choices across projects and the main determinants of that variability. This analysis is valuable because a firm that manages multiple projects can benefit from the careful analysis and consideration of the interactions among and the specificities of its projects. Using a comprehensive database of construction firms and projects, we conclude that firms demonstrate variability in their insource or outsource choices across projects and that this variability is explained by factors such as the number of projects simultaneously undertaken, the variability in a project's complexity, and their market power in local markets. These results suggest that the theories explaining firm boundaries in project-based firms should be expanded to include interrelationships between projects and the individual project characteristics that drive differences in insource or outsource choices.  相似文献

The contribution of fecal pollution from dogs in urbanized areas can be significant and is an often underestimated problem. Microbial source tracking methods (MST) utilizing quantitative PCR of dog-associated gene sequences encoding 16S rRNA of Bacteroidales are a useful tool to estimate these contributions. However, data about the performance of available assays are scarce. The results of a multi-laboratory study testing two assays for the determination of dog-associated Bacteroidales (DogBact and BacCan-UCD) on 64 single and mixed fecal source samples created from pooled fecal samples collected in California are presented here. Standardization of qPCR data treatment lowered inter-laboratory variability of sensitivity and specificity results. Both assays exhibited 100% sensitivity. Normalization methods are presented that eliminated random and confirmed non-target responses. The combination of standardized qPCR data treatment, use of normalization via a non-target specific Bacteroidales assay (GenBac3), and application of threshold criteria improved the calculated specificity significantly for both assays. Such measures would reasonably improve MST data interpretation not only for canine-associated assays, but for all qPCR assays used in identifying and monitoring fecal pollution in the environment.  相似文献

Here we report results from a multi-laboratory (n = 11) evaluation of four different PCR methods targeting the 16S rRNA gene of Catellicoccus marimammalium originally developed to detect gull fecal contamination in coastal environments. The methods included a conventional end-point PCR method, a SYBR® Green qPCR method, and two TaqMan® qPCR methods. Different techniques for data normalization and analysis were tested. Data analysis methods had a pronounced impact on assay sensitivity and specificity calculations. Across-laboratory standardization of metrics including the lower limit of quantification (LLOQ), target detected but not quantifiable (DNQ), and target not detected (ND) significantly improved results compared to results submitted by individual laboratories prior to definition standardization. The unit of measure used for data normalization also had a pronounced effect on measured assay performance. Data normalization to DNA mass improved quantitative method performance as compared to enterococcus normalization. The MST methods tested here were originally designed for gulls but were found in this study to also detect feces from other birds, particularly feces composited from pigeons. Sequencing efforts showed that some pigeon feces from California contained sequences similar to C. marimammalium found in gull feces. These data suggest that the prevalence, geographic scope, and ecology of C. marimammalium in host birds other than gulls require further investigation. This study represents an important first step in the multi-laboratory assessment of these methods and highlights the need to broaden and standardize additional evaluations, including environmentally relevant target concentrations in ambient waters from diverse geographic regions.  相似文献
